You need to have a full time job and you have to apply with Student Pass first and once you got the job the company would sponsor the "employment pass".
Once you've the employment pass then you need to apply for permanent residency.
So it's not like you get EP first and then apply for a job, it's the opposite.

No such thing as "valuable" but it's based on your preference and priority. MBA - I wouldnt recommend for a fresh graduate. MBA helps when you've lots of experience but need a systematic way to streamline your working with industry/academic standards.

MCA - Typically it's Indian course and not offered in Singapore.

MS - This is nothing but Master of Science or MSc which is offered in India. Also note that even if you do MSc (or M.S) from Singapore universities your pay (may be the 'value') will be lower than B.Tech graduates, just like in India.
